Prediction of Hyoscine from Amethyst Fruit (Datura metel L.) as Beta-adrenergic Receptor Kinase Inhibitor for Heart Failure

Hyoscine, suatu alkaloid tropan yang diperoleh dari Datura metel L., telah lama dikenal karena aktivitas farmakologinya sebagai agen antikolinergik. Studi ini bertujuan untuk mengevaluasi potensi hyoscine sebagai inhibitor β-adrenergic receptor kinase (β-ARK) melalui pendekatan in silico menggunakan Prediction of Activity Spectra for Substances (PASS) online. Hasil prediksi menunjukkan bahwa hyoscine memiliki probabilitas aktivitas (Pa) sebesar 0,952 sebagai inhibitor β-ARK, sementara probabilitas ketidakaktifan (Pi) hanya 0,002. Nilai Pa yang mendekati 1 menunjukkan tingkat kepercayaan yang tinggi terhadap potensi aktivitas hyoscine dalam menghambat β-ARK, suatu enzim yang berperan dalam desensitisasi reseptor β-adrenergik. Penghambatan β-ARK berpotensi meningkatkan respons reseptor terhadap agonis β-adrenergik, yang dapat dimanfaatkan dalam terapi kondisi kardiovaskular dan gangguan sistem saraf. Temuan ini memberikan dasar awal untuk pengembangan lebih lanjut melalui studi eksperimental guna mengonfirmasi aktivitas biologis hyoscine terhadap β-ARK.
