Abstract

Hyoscine, a tropane alkaloid obtained from Datura metel L., has long been recognized for its pharmacological activity as an anticholinergic agent. This study aims to evaluate the potential of hyoscine as a β-adrenergic receptor kinase (β-ARK) inhibitor through an in silico approach using online Prediction of Activity Spectra for Substances (PASS). The prediction results show that hyoscine has an activity probability (Pa) of 0.952 as a β-ARK inhibitor, while the inactivity probability (Pi) is only 0.002. A Pa value close to 1 indicates a high level of confidence in the potential activity of hyoscine in inhibiting β-ARK, an enzyme that plays a role in desensitizing β-adrenergic receptors. Inhibition of β-ARK could potentially enhance receptor responsiveness to β-adrenergic agonists, which could be utilized in the therapy of cardiovascular conditions and nervous system disorders. These findings provide an initial basis for further development through experimental studies to confirm the biological activity of hyoscine against β-ARK.
